1. 2009 Resident Research Grant Award from CAP Foundation (10/01/2009 – 11/01/2010) Title of Project: GCET2 Expression in Acute Lymphoblastic Leukemia/Lymphoma |

Summary: GCET2/HGAL is one of several germinal center (GC) B-cell genes that I cloned during my PhD training. It is a membrane adaptor that, upon B-cell activation, recruits cytoplasmic proteins to the cell membrane to form a signal transducing scaffold. GCET2 is one of the top markers for normal GC B-cell and GC B-cell derived lymphomas. We also found that GCET2 mRNA is highly expressed in the thymus and immature B-cell lines. The major purpose of this study is to investigate GCET2 protein expression in clinical cases of acute lymphoblastic leukemia/lymphoma, and the protein expression will be correlated with clinical behaviors, cytogenetic aberrances, and prognoses in the clinical samples.

2. Virtual Karyotyping of Chronic Lymphocytic Leukemia (CLL) by SNP Array |

Summary: SNP arrays provide a high-resolution “virtual karyotype”. Our data have shown that SNP array is more sensitive in detecting genomic aberrances in CLL than conventional karyotyping and FISH. This project specifically focuses on subclassification of del(13q14) and copy neutral loss-of-heterozygosity of 17p (CN-LOH 17p). Del(13q14) is heterogeneous and can be subclassified based on length of deletion and deletion of single or double alleles. The subtypes of del(13q14) are associated with different clinical behaviors of CLL. CN-LOH is important in tumor pathogenesis, in which the tumor may acquire two mutated alleles of a gene from one parent. |

3. Classification of Diffuse Large B-Cell Lymphoma Using the New Algorithm (Current) |

Zifen Gao Summary: Diffuse large B-cell lymphoma (DLBCL) can be divided into two major subgroups by gene expression profiling, germinal center B-cell-like (GCB), and activated B-cell-like (ABC). GCB-DLBCL has a significantly better prognosis than the ABC group. Recently a new algorithm was proposed using five antibodies (GCET1, CD10, BCL6, MUM1 and FOXP1) to classify DLBCL into GCB and non-GCB subgroups, which showed higher sensitivity and specificity than the previous Hans algorithm.

4. Thymidylate Synthetase Expression, Genotype and Classification in Pseudomyxoma Peritonei (2007) |

Summary: Pseudomyxoma peritonei (PMP) is characterized by peritoneal spread of a mucinous neoplasm, most commonly of appendiceal origin, and massive accumulation of mucin in the abdomen. PMP has two histologic and clinical subtypes: low grade (disseminated peritoneal adenomucinosis, DPAM), and high grade (peritoneal mucinous carcinomatosis, PMCA). We evaluated the expression of thymidylate synthase (TS) in PMP and its correlation with tumor grade, proliferative fraction and expression of p53. We concluded that TS expression is significantly higher in high grade PMP than in low grade PMP, and TS expression correlates with p53 and especially Ki67. These findings are consistent with the TS data on colorectal cancers

5. Cloning of Germinal Center B-Cell Associated Genes, GCET1 and GCET2 (2000-2006, PhD thesis) |

Summary: DLBCL has been divided into two major subgroups by gene expression profiling, GCB and ABC. Many genes in the GCB-DLBCL have not been cloned and are represented as short expressed sequence tags (ESTs). The goal of my PhD thesis was to clone and characterize several GC B-cell associated genes from selected top ESTs in the GCB-DLBCL. Two new genes, GCET1/Centerin and GCET2/HGAL, were cloned using the rapid amplification of cDNA end (RACE) technique. Both GCET1 and GCET2 are among the top markers of normal germinal center (GC) B-cells and GC B-cell derived lymphomas. GCET1 belongs to the SERPIN family. GCET2 is a membrane adapter that can be phosphorylated by protein tyrosine kinase, Lyn and Syk. Phosphorylated GCET2 recruits cytoplasmic SH2 proteins to cytoplasmic membrane to form a signal transducing scaffold.

6. Classification of Diffuse Large B-Cell Lymphoma (2003) |

Key investigators: Dr. Christine P Hans and Dr. Wing-Chung (John) Chan Summary: In the project, an immunohistochemical stain algorithm (the Hans algorithm) with three antibodies (CD10, BCL6 and MUM1) was used to classify DLBCLs into GCB and non-GCB subtypes. It resulted in ~ 80% concordance with gene expressing profiling.

7. Clinicopathologic Features, Immunophenotype and Epstein-Barr virus in Nasopharyngeal Lymphoma (1996-1997, medical school thesis project) |

Summary: 112 cases of primary nasopharyngeal lymphoma from Northern China were colleted to study the clinicopathologic features, immunophenotype and Epstein-Barr virus (EBV) infection of these cases. 77/112 cases (68.8%) were T-cell lymphomas, and 35 cases (31.2%) were B cell lymphomas. EBV infection was identified in 54/101 (53.5%) cases, including 48 T-cell lymphomas and 6 B-cell lymphoma, by EBER in situ hybridization. |
